After I have played couple hours of the beta, my pinky feels a bit hurt because I had to use it to hold down the shift key most of the time, especially playing range class like demon hunter. I wonder if anybody else has the same issue. Also please post your key bindings if you are using one which is different from the default.
I've been using Shift for Hold. Alt/Com + lft and rght mouse buttons for 3,4,5,6. It works ok until I have to Hold then it gets confusig. might try Space and a,s,d,f.
Only thing I've changed so far is.... SpaceBar = Health Potion, I play hardcore so I wanted something big and easy to hit quickly if need be. The next thing I changed was I made the Mouse Wheel a skill slot. The reasoning was, I can put left click as my main generator, right click as an AOE that I use often such as arcane orb, and then the mouse wheel is either single target high damage or something that's on cooldown AOE such as frost nova or soul harvest that I still use alot but has a cooldown.
I'm gonna remap Shift to Spacebar, sounds awesome (especially considering I played DH all morning today. With a touchpad).
Got a two extra button mouse so:
I have potions as my "oh shit!" button on mouse 5. Actually feels a bit awkward, pretty easy to use on accident, but I think that's actually a good thing... "fuck fuck fuck" "Oh, potion" (I intend to play HC)
I got the "move" command on mouse 4. Not sure if I'll be using it, though. May put something else there instead. Those lore popups are annoying.

Anyway, since you don't need SPACE in this game, you could probably swap it with LSHIFT
default
Spacebar for hold is a good idea though, will try that.
Mouse1: primary attack
Mouse2: secondary attack
Mouse4: potion
Mouse5: shift
1-4 skills stayed as the default, on the keyboard
Nice one with all the skills binding to the mouse. I'll try that.
I would recommend one to anyone playing moba's or mmo's that aren't fps.
