I think the horsemen could be a fun side quest...it may not be completely original. However, it is somethign a lot of people would notice and relate to. Which is indeed fun even if it seems gimmicky.
If I were to take on this sort of side quest task, using red/black/yellow/white deaths. Instead of knights, I think they could be represented with some pretty ferocious and unique demons. One could be massive and completely destroy its surrounds because of how out of control it is...almost as if it were overloading and not even aware of whats happening. Another could be extremely strategic and pick the player apart using its skillsets in a very efficient manner. One could even be pretty big at first, but break apart into three different parts, each with its own stats....ie: agility, defensive, offensive, magical, melee, etc. Just for fun one could be a complete dimwhit, get beta down and give up the locations of the others in the form of riddles...adding more to the side quest..having to solve a mystery of sorts just to finish.
Those are just ideas, nothing to take as if it will ever happen. Just trying to think outside of the box a tiny bit. But trying to give players something they might be able to relate with.
i like the 4 horseman idea. it reminds me of Zelda for some odd reason though.
anyways, anyone heard of a 'wendigo?' its native american myth. i dont know if it is boss-worthy, but the opening of this page is really cool: http://en.wikipedia.org/wiki/Wendigo

and since there seems to be an asian area, why not an asian themed boss? any ideas?

Yeah, the Wendigos weren't that threatening... but Treehead Woodfist was. So fast, so strong. Scary when you're only level 6. Thats such a cool name too.
I want to see all those class from D2 come back as bosses
The Barb guy returned to the cause as we can see and the Paladin guy became the king of Caldeum. The rest I don't know about. I heard a rumour that the Necro guy will make a cameo as an NPC. Something like that was hinted anyway. There's probably going to be a quest or two relating to the old classes.

I agree that this would definitely be a Random sidequest chain. Having them be demons instead of horsemen is original.

Personally, I'd hate that. We have enough demons already. The whole damn game is demons-demons-demons. Give us something original for once.
5 things we could have that suit the Diablo universe:
1. Demons (original, eh?)
2. Undead (even more original, i know)
3. Humanoids (bandits, cultists, that kind of stuff)
4. Beasts (any kind of animal, natural predators or simply twisted by the powers of Hell)
5. Angels
nr 5 is the only thing we havent had yet. The novels talk about how strict the powers of the High Heavens are and how in their struggle with the Burning Hells they'd most likely destroy Sanctuary instead saving it. - that could be what we need to actually fight them. But only as bosses because their powers are quite supernatural.
I'd go with some side-quests about some humans driven mad/gone evil - could be heroes of Diablo 2, or simply lore characters and with animals (as for originality). I mean, we havent had any quest about the local wolves twisted by demonic forces or who knows what else, that might lead us to different bosses.
There are also quite a few legends about animal monsters in every mythology in the world, i'm sure there could be some inspiration.
I want to see the other Seven Sins incarnate (Thousand Pouner is 'Gluttony Incarnate'). I always enjoy such features.
I'd love to see a boss that makes use of the 'light circle', leaping in fast, dealing out 1, 2, 3 hits then slipping away again. Like a compact, humanoid exoskeletal demon thing. Black, of course.
I want to see angels! I think we will anyway... maybe in the expac if not the main game.
More enemy mage bosses! That do more than just stand there casting missile spells (I'm looking at you, Summoner, Lazarus). They should have unique abilities too (that may be usable through certain items :D)
And I like the horsemen idea. But I think it should be Pestilence: Conquest is too similar to War. And it would be more fun to fight them all at once, as has been suggested.
Pestilence is more commonly known, so sure.
The seven Sins incarnate is actually an awesome idea. What were they again? (I'm afraid I'm not much of a sinner). Gluttony, Lust, Pride, Envy........ and I'm lost (the others are probably some of my weaknesses). Anyone wanna help me remember?
could be a sweet boss...if the horror level was raised. For example, if it was hard as hell to track it, see it, and if it had some downright nasty abilities.
lust could be a beautiful being that is deadly in some sort of sexual way btw Asmodai is the demon of lust
greed could be something like a king midas like character, who has great drops after you off him
sloth is laziness and or depression, dunno what would fit, but dementors from harry potter somehow flash across my mind
envy should probably be story related because it has to do with certain qualities of a person, or their material wealth
pride is probably conveyed by one of the lesser evils, or a 'competitor' of the highest powers
seems like a fun idea.

If we had horsemen, men on horses, I think there would be a discrepancy with the fact that you cannot have mounts and NPC's can.

those dont even look nearly as threatening, you must agree! i always called those things brown yeti's so that explains something.
what about a spider boss, you have to take out its limbs before attacking the main body? giant snake would be really fun too
