Based on the Black Soulstone cinematic, I'm pretty sure Deckard Cain is going to die, probably in the first act.
Leah is sitting there pouring over his texts, trying to figure something out, verbally asking her uncle what she's missing... she's talking aloud to someone that's not there. He could have just been captured, but I think it's pretty clear he's dead.
This is probably (one of) the spoiler(s) that Metzen mentioned was cut out of the cinematic.
I think it's a bold, gripping move to pull out in Act I though, to kill off the only character that has been with us since Diablo I. I think it's a good way to open the story up after reintroducing him, pulling the player in, and giving us that "NO!" moment when he falls.
I think this is why they brought Leah into the story... to replace the fallen Deckard Cain.
If you've played the Beta, you know she doesn't really believe in the prophecies that Deckard Cain does, and I think it's the perfect setup, for the elder to die and the non-believing youth to realize he may have been right, and it's up to her now to figure out what he knew.
Edit: Hm... on the main page, they're saying the black soulstone cinematic is THE opening cinematic for Diablo 3. If this is true, then Cain is probably just trapped in the cathedral, but I'm guessing this is the end of Act I cinematic, after you've discovered the black soulstone, and the opening cinematic will have more to do with the fallen star.
Well, the Wanderer was possessed by Diablo before. Why Leah couldn't be possessed by Diablo this time?
Azmodan cannot posses anyone because he is Fat, so he simply doesn't fit xD
I think this is actually incredibly plausible. Why else would she act that way? After reading the description of Belial, it does seem that he could have been Cain the entire time.

ive been thinking this for a while now, what if Cain is behind the meteors fall? being one of only 2 people to survive its impact the other being Leah makes me think he had a role in it all

By "All" he may mean the prime evils, and he may be talking to the Horadrim in general, reflecting back on how the Horadrim captured the Prime Evils; as based on lore, Deckard is attempting to teach Leah about the tasks the Horadrim are responsible for, considering he is becoming quite old...
As for Deckard Cain and how she calls out "what am I missing, Uncle?"... I don't think Deckard Cain is dead, but simply absent (I.e. In bed because it's late? And she's doing some late night study?)
Someone above also mentioned that Deckard is Belial... That wouldn't make sense since Belial appears in Act II, and I'm speculating that Belial is actually Haken's son, Haken II. Haken is the leader of Caldeum and dies, but his son is too young to rule, so He is guarded by Asheara (diablo II, Act III mercenery seller) until he becomes of age but he tells them to stand down and wishes his own guards to protect him (showing that he may have been corrupted by Belial)

That seems rather far-fetched however. The Horadrim didn't mastermind no elaborate plan to fool the Great Evils, they were just "Holy crap, there's three Prime Evils in our back yard! What do we do?" until Tyrael dropped down and helped them with the soulstones. Specifically when you account for the "I defy you" part, it's doesn't make much sense to say that to some random chick who happens to be the adopted daughter of the last Horadrim.

Maybe, but why not just wait for him to come back? She seems rather distraught with not being able to figure it out herself. I get the feeling it's more than him just being temporarily absent.
PlugY for Diablo II allows you to reset skills and stats, transfer items between characters in singleplayer, obtain all ladder runewords and do all Uberquests while offline. It is the only way to do all of the above. Please use it.
