Well, the last few seconds of the speak peek video confirmed my thoughts about how Diablo can be alive after his soulstone was destroyed and why he looks like a female.
SPOILERS!!! (From the Beta and maybe potential future gameplay if I'm right)
I believe that since Leah is Aiden (warrior from D1) and Adria's child, that a part of Diablo's soul was transferred from a Diablo Possessed Aiden to Adria's now impregnated egg. The Beta lore from Adria said that they spent the night together after he returned from the Cathedral. With his soulstone destroyed, Diablo was supposed to die, but his spirit slowly gained strength again within Leah. I believe this is why she dreams about Azmodan in the Blizzcon cinematic (remember Marius saying "Why do things always happen as they do in dreams?" and him dreaming about Baal while in the presence of Aiden/Diablo).
Nevertheless, I believe this is why Diablo is now in a female body. Leah carried him as her father did.
