I would like if they sharpened the graphics a little and added more textures, like grittier and bloodier stone. Yes, cartoony = bad.
However, I disagree with making the game more gray. Gray = more boring imo, and more of a sore to my eyes. I like the color.
You can still have spookiness and gore with a color.
I loved the brief conversation with Cain. The nostalgia from "Stay a while, and" was great, and "Bah, nobody ever listens" was hilarious.
Also, I think the girl's voice actor (who also voice acted Sylvanas in Warcraft) is great.

What will the skill system be like?
Will there be sets?
Will there be the Horadric Cube, or something similar?
Will there be runewords?
Will there be uniques?
Will there be any shapeshifters (werewolf, werebear, somebody who can turn into a demon, etc.)?

On the second comparison, I prefer the original. I like the color.
Yeah, yeah, D1 was scary, blah blah... but none of the games actually evoked heart wrenching fear. The Diablo games aren't horror movies; they're just games with a dark atmosphere. I enjoy the dark atmosphere, but I also enjoy the lighter fantasy element. With the first comparison, I agree, it should look a bit bloodier and darker - you're in the spooooky dungeons. But outside, I want some fresh air along with my monster slaying. Besides, I'm sure there's going to be plenty of other outside environments that are darker, like jungles, icy places, or anywhere nearer the bigger Bad Guys (I say Bad Guys 'cause we don't know if all the Prime Evils are returning, do we?)

More blood good, better textures on stone surfaces good, rest... *shrug* The gameplay video had a few darker corridors, so I don't think the grayer tone is needed... but it does look cool. Maybe there could be an alternate graphic setting? lol... like there's a vivid, intense, and couple other graphic settings in UT3

I liked the druid too.
I really hope there's either a druid style wolf/bear shapeshifter, or a demon shapeshifter, like Illidan in W3.
I'm upset by almost everybody automatically assuming that there's going to be a paladin.

Barbarian looks like he's going to be a shitload of fun, leaping and charging around. Charge and shouts will probably replace D2's paladin.

Witch Doctor also has fire and ice, which makes him a bit like the sorceress/druid/necromancer mix, which is a lot.

I think they've got to have a more mainstream magic caster than the Witch Doctor, so a sorcerer/elementalist seems pretty likely.

Someone who is proficient with a bow is a must-have. In D2 that was the amazon, but now that we've got female barbarians, I think the amazon isn't as likely as a new archer/assassin/thief mix. (the rogue in D1 was basically just an a bow/knife wielder)

That would leave one class left. Maybe...
A more healing oriented paladin (I'm hoping for something weirder)
A druid/hunter mix (only problem is this would be a bit like an Anti-Witch Doctor - but I'd love to play as a Werewolf again, or to be able to summon puppies to horribly maul my enemies)
A demon and curse oriented Necromancer (Necro of course kicks ass, and I think they could make the class sufficiently different from the Witch Doctor)
However, my bet goes to this:
A class that uses powers directly from demons and angels, and can even summon lesser demons and angels (I think this class would have a lot of potential - summon lightning from heavens, summon zergling like demons from the earth, and transform himself into a powerful being, Illidan style)
