yes, if one takes a look at the map of the world of sanctuary, you will see that there are several areas that we have not been to yet.
-the entire kingdom of westmarch, (where the paladins now come from, they used to come from Kurast, read the history)
-South of the Temple of Mephisto and his Durance (Prison)
-North and East of Kurast where the sorcerer, assassin, sorceress and necromancers come from
The Amazon Homelands
-East of Harrogath, Scosglen home of the Druids,
so it is possible for at least one of these settings (well probably not the amazon/druid homelands) to be an asia setting,
act 1/5 = Europe

Asia (and Heaven), is about the only setting missing, so it is possible for Samurais and Ninjas to exist somewhere within the world of Sanctuary.
i think they are very doable.

well diablo has influences of Europe, Middle East, and Latin America, so why not Japan, its one of the only medieval settings not yet used in diablo

i think, both a ninja and a samurai can be done in d3,
samurai would have a lot of sword techniques, critical strike, called shot, power shot, defensive/offensive spin, and a few other magic skills, spirit armor, magic armor, enchant blade
ninja wouls have some sword techniques, sneak attack, a lot of ranged techniques, magic shurriken (similar to the necro skill teeth), and stealth techniques like hide in shadows

i have to say this, but ninja and samurai are very different,
Samurai, were the knights of japan, fighting wars for the shoguns, protecting villages, while ninja were assassins, hired mercenaries, and later bodyguards of tokugawa
samurai have more incommon with the Paly/barb while ninja have more incommon with the assassins, naturally
