I'm pretty sure PCIe Rev 1 wont work with a PCIe Rev 3 card. If it does It most likely will have no performance issues although I can't be sure considering that we don't have any info on that. I mean PCIe Rev 2.0 isn't used to its potential either so I'm almost positive you mgiht be able to run it. Not many people use PCIe Rev 1 anymore most upgrade to 2.0, If you're still running a board that old though i think it's about time for a upgrade. What are your specs?

My specs are:
i7 920 stock
12 GB RAM (using cpuz, under memory, it says DRAM speed is 533MHz, under SPD tab, it says 667 MHz, which is it, 1066 or 1333?)
TBGM01 mobo ... (ewww...)
750W PSU (FSP750-80GLN)
OC'ed GTX 260
If i were to upgrade my mobo, Id might as well upgrade everything, but im trying to delay for another 2 years, and just upgrade to a better card for now. This rig has lasted me 2 years, and I think a new card would allow me to run what I would like for another 2 years.
Your motherboard is PCI Express 2.0. Compatibility shouldn't be a problem.
In fact, the 260 video card is probably enough for high details in Diablo 3 at 1080p-or-less resolution. You might have to turn shadows off, but that's it.
-Your mobo is pci-e 2.0, not even close to a bottle neck.
-Your 920 @ stock is still overkill for this game.
-GTX260 is also plenty for this game, but if you must, sure a GTX680 wont break a sweat.
Since the 680 is the only one out yet, lets take it as an example..
would performance be affected by using a PCI-e 1.0 16x slot on the motherboard ?
would it be noticeably bottlenecked by an i7 920 stock ?
All these troubles because of a crappy mobo with pcie 1.0 and no overclocking cpu function...sigh

You're running an I7-920. That's socket 1366, which uses the X58 chipset, which is all pci-e 2.0. PCI-E 1.0 has been gone since the end of 2006.
