Definitely vanity gear should be on sale, since if someone wants to be luminous Red, with glowing blue shoes, and spikes coming outta their back, they should be able to purchase that for effect (PvE only - Would give an unfair PvP advantage)
I also don't mind Blizzard item stores - To an extent.
The following is D2 related, but equivalently applies to D3 as well.
If they sold the equivalent of Lower / Mid Lvl D2 sets (Isenheart / Natalya sets, not maxed Tal'Rasha) / Uniques (Sure let them buy a Exceptional Unique's - just not Elite Uniques) / Runes (If people want to pay for a Pul - Let them - But don't let them pay for a Jah / Cham )- Then sure - Let people buy them.
A system where people can pay for mid-level gear I'm fine with. It will let the "Casual" gamers shell out cash if they really want to get strong quickly, but still let the hard-core fans have the uber stuff!
Tempted to say vanity items only, but I saw what happened in Team Fortress (the store is now full of items that affect playstyle, even though Valve said this would never happen). So I want to leave that door closed forever and keep Diablo's integrity. I'm fine with people paying for more player slots, renaming characters, or even complete respecs (different from the ones you can buy with ingame gold).
Casual players should not be able to spend a few bucks and instantly get the gear that the hardcore spent hours trying to get. The economy should only be within the game itself... outsourcing the economy basically makes all in-game trading worthless.
I voted Blizzard store to begin with, but then I changed my vote to no stores.
I do agree that players shouldn't be able to buy items to basically shoot their way up to a prestige that they didn't earn by playing, I think we can all agree to that. The same goes for unique things like visuals, dyes, stuff like that.

Items are called loot in this game for a reason. They are your bounty, your incentive to go out there and fight under the risk of getting killed. Sure the quests and XP are nice and all but we all know the big part of the Diablo experiencie is the loot.
I voted vanity items, but only for PvP. Like someone said, special taunt when you win and stuff like that. Nothing more. There is absolutely no reason or the need for stores. The only thing necessary to acquire gear in D3 has to be time and skill.
No stores in my opinion, if you don't work to get something, you don't really appreciate it either.
Seeing as Blizzard aims to use in game gold as the main currency, I doubt there will be much item trading and the need for sites as d2jsp etc. However, goldfarming and gold selling sites will most likely be a fact a few months after release, and unless they find an effective way of eliminating/punishing these offenses, some people will always "cheat" their way to the top, but that's their loss.

I honestly don't think bottin is going to be much a problem with the security bnet 2.0 should have, but i do worry about 3rd party sites like d2jsp where one could just buy Forum Gold with money and buy gear from other people without having to put in any effort at all... I just hope blizzard tells them that's a big NO NO.

Everyone will know that they paid for it, so its not like they'll somehow be better or anything, and it gets Blizzard more money to improve the game with. Seems perfectly fine to me. Simply put - if you can buy the item, then there is no worth to it - speaking from a luck factor of finding loot in D3. Whether you play 80 hours a week or 2, has no relevance. The thrill of loot is finding it, not buying it.
Granted buying it makes you excited to play again, because you are godly the moment your card clears. However, that's a cheap thrill and ultimately not in the spirit of this game.
I would personally prefer if there were no item stores at all, from any source, in any game, ever. However, given the obvious attraction that most companies have to them now, and the fact that gamers pump money into them regularly, added to the fact that WoW already has some cash items for sale... we can assume there will be some sort of sale of items in Diablo 3. Even if it was impossible for third parties to do so, which I seriously doubt will be the case, we will likely see promo and vanity items like the ones in WoW for sale. So while I voted for vanity items, because I think that is honestly a best case, real world scenario, I wish that there would be no cash sales at all.
All the same, clearly a lot of people like the idea of being able to buy things that offer them no real use or benefit, so more power to them and companies that cash in on them, I suppose.
For me User-hosted Stores workz best because if someone puts effort in farming the gear than they desrve somekind a reward.
But still you cant be good at the game only if you buy ultra cool super-powerfull stuff , you need to play trough it to see and to experience stuff and you need to devote lot of time just to gain some essential and all that...

If you find an item thats worth somthing to another player selling it for gold or trading it for an item worth somthing to you should be rewarding enough,If by User-hosted store you mean like a trading post system,then sure but for a player to find an item ans sell it for real money to the highest bidder seems unfair and unwise.
I think it's also important to point out that the sales of in-game or otherwise virtual items are considered illegal in some places, as well as simply violating the ToS of many games as well as selling sites such as eBay.
I'm gonna have to say I lean to the no stores side of things, but with that said I also have to admit that there is going to eventually be some site selling gold and maybe items. As long as Blizzard does its job which they sounded pretty adamant about making a strong economy, then we will be fine. Any thing that is bought will be straight from the economy and just as easily (or difficultly) earned by putting time into the game. Sure there may be a character or two out there that was buying some pretty good items but it's still something that had to drop in game and put up on the market. It's never gonna be the only way to get the best items so haha to those who spent money. Blizzard making a store and essentially duping items for distribution is the dumbest thing I ever heard. That would go against the economy they are fighting for and that needed to be said.

How can there be a best non-useful item? If there are 5 vanity dyes, who but the individual can say which one is best?
The vanity you can get through purchasing items is not the same as the one you would get for, say, wearing a full super rare set. Regardless of how special you look with custom auras, glowing hands, golden armor, etc, everyone will know that those are all vanity items purchased through a store, not gained through hours of monster slaying.
Any item that is difficult to get is more useful. If one of the dyes has a 0.001% chance of dropping off of the end boss, then it's obviously going to be worth more.

Store items can bind to the character or the account. Problem solved.
Hah, problem not solved. Person X just bought the $5 dye from the Blizzard store. But person X was the person who was going to buy some other dye from me for in-game gold.
