There is a lot they could explore. The mage wars, the original imprisoning of the 3, some new demon that claims throne of hell, actually play as angels and demons in the eternal conflict independent of sanctuary, an. Mmo might have potential, the time period in which Ureh was this beacon against hello. Many of these would require fleshing out lore and some retcons I am sure.
Caldeum is officially confirmed I think for being a major place in Act 2. If not officially confirmed then absolutely 100% proven through datamining.
I'm not that concerned with how the plot will evolve during the game because the majority of people in the lore section have already figured out about 60% of it. What I'm curious about is what will Blizz do to open the doors for the 2 expansions that follow.
Diablo escapes? Not likely. One for Mephisto and one for Baal - more likely. After seeing Diablo for 5 years now through official and fan art and tons of lore speculation... I'd like to hear a lot more about the other two now.

My guess is that there will be some hint, if not them pretty much clearly stating, what the next expansion will involve.

I think you should start with lore by going over by our wiki. Don't take it the wrong way but you'll learn very, very interesting things about the creatures you mentioned and our wiki team + some people like Doomscream have made it so that we know absolutely everything that has slipped off of Blizz's lips.
You won't be disappointed and you'll get into what's really going on in D3. :]
I know it may sound silly, but I can't recall being this excited about anything for years. D1, and D2 represent some of my fondest gaming memories which themselves were a major part of my youth. It feels like I am about to reunite with an old friend and we have LOTS of catching up to do!

I bet heaven and hell come together against nephalem in the final installment and the eternal conflict is ended? I mean what else more cataclysmic could there be. I think it's pretty obvious that we save what we can of heaven in launch a4. Maybe faction disputes will occur between nephalem again as uldyssian-like ldeaders rise up and begin taking different sides-with heaven or hell. Who really knows. Despite knowing many of the dots and maybe all of the major players, I can only connect so many for the pre-expansion content.
