Diablo for sure. You can't have a sequel without the man himself. I would assume that Mephisto and Baal will make a reappearance. Baal caused more chaos than Diablo himself did, so I would assume he is back. Mephisto created a lot of turmoil in the east and he is extremely bad ass, so I would figure he will be back too.
I would also assume all four Lesser Evils will be included: Andariel, Duriel, Azmodan, Belial. The game will be much bigger, and can handle them all.
It is almost a certain guarantee. Blizzard is known for their very detailed lore and beckstory in all of their games. They all have their own history that is very detailed.
In the center of this history stands the conflict between Heaven and Hell, and the leaders for Hell are the greater and lesse evils. If we were to remove them, the threat from Hel would be greatly reduced, and the conflict is pretty much over, and the good guys win.
This leaves us two options if D3 is a sequel:
1. Diablo and possibl his brothers, Andariel and Duriel are brought back from the dead. This could be done by a mortal agency, by one of the two remaining lesser evils, or some natural occurence where they are spontaneously respawned. No one knows.
2. The title of a Prime Evil is not static, and so could change. WE do not know how the ranks in Hell work, is could be that when a Prime Evil falls, a lesser evil will step up. Or there are other evils and ther planes of Hell that we have yet to see which hold other demons.

I think diablos father is going to be apart of diablos 3 storyline, he is going to do some ritual where he needs the blood of an archangel (tyreal) to bring all the dead sons back. They somehow kill tyreal ( sad ) and baal.meph and diablo get all ressurected back to life. MY STORY>> WOOT
I hope there is no Diablo's "father" or "mother" or "cousin-in-law". I want the Prime Evils to be standalone beings. No father or mother - just born from chaos.

I doubt that will happen. I see the term "brothers" applied more on the relationship between their domains (Hatred, Terror and Destructon) rather than their actual selves.

I seriously hope that Diablo comes back but the others I can do without. I'd really like to see what all hell has hidden from our eyes, what if the greater evils man knows about aren't even the surface of the true depts of Hell?

we need the Dante's Inferno Hell. the 9 layers of torment. or at least one that is as vivid and scary.
though the gothic architecture of the Chaos Sanctuary was nice though. but it didnt seem as scary as levels 13-16 in diablo with the giant bone walls and implaed/decapitated/mutilated corpses everwhere.
but to answer the question "will there be any old bosses be in d3" well Diablo of course. and depending on the manner of his return, i would be surprised to see 1 or even both of his brothers (not his actually blood brothers but more like brothers in crime so to speak) Mephisto and Baal. As both Andariel and Duriel are lesser evils they can be much more easily revived than the Prime Evils, so seeing either or both of these two again is not completely out of the question.
in d1 you had The Butcher, in d2 you had The Smith and The Armorer, so i d3 i would expect there to be at least one demon with the name of a profession.

My fav is Mephisto then Diablo then Baal, baal looks like a fag, mostly in the cinematic were he blows up that bald dude.
