I used to run, often, in highschool, as conditioning for my martial arts training.
No problems back then.
Then I let myself get really out of shape, over the following years (up to 310 pounds, in late-2004).
By Dec. '05, I had brought myself down to 192 (5'9). I tried running again at that point, but after a few sessions of interval-sprints, I had to stop because I could barely walk -- knees were killing me.
All the men in my family seem to develop knee problems later in life. My father is a walking (hobbling) pain-medication receptacle. My knees aren't as bad as his, but I decided not to push my luck.
I do fine with my bike.

Did you have a doctor look at your knees, read material about it? That sounds serious.
I get knee pain while running for the first time, but it goes away after some time.

But they only hurt if I run -- no pain at all when I walk, or bike; nor when I place them under a heavy load, as when doing squats.
I figure they just can't take the high-impact at the weight I am these days (was 135-150 in highschool).
Perhaps they would adjust over time, if I was careful. I just haven't made the effort to find out, since I have other, equally enjoyable (for me) options.

You are never supposed to run until things hurt. There should be no pain involved in or after running.
Take a biology and anatomy class and don't say stupid shit like that again. What you said only applies to trained bodies. Everyone else will encounter pain in the following areas:

I do think it is simply, like you said, that I have not re-acclimated my knees since getting back into shape, so maybe my initial statement of "can't take it anymore" was a bit too extreme.

That's stupid shit and I don't advise you to be giving out shitty advice.
If you feel pain while running, stop. Rest until it goes away, and then run that SAME amount again. If you don't feel pain, good. If you do, stop and rest. Repeat until there is no pain for that distance. Slowly increase how far you run each time if no pain is present. Note that by rest, I mean over night, or a couple days, not a couple hours.

I am in pretty good shape, and I rarely do any exercise. Example: I randomly walked 7 miles both ways and didn't feel anything. (Granted I run out of breath and get pains in my side after running half a mile, but if I could learn to pace myself then I could run a whole mile without stopping..)
From what I understand, (And excluding the bullshit alot of these fitness centers and fitness videos shove down your throat to make another buck) everyone has a certain plateau that they can reach, and some people are naturally fat/skinny/fit. But I am not completely certain about that.
