I am categorizing facts we know that will directly effect the story of Diablo 3. Hopefully having the information in one place will make it easier to theorize. If anyone can add anything please do.
D1/D2:
Diablo was defeated and his soul-stone destroyed. Same goes for Mephisto, and assumed for Baal. All three SHOULD be in the black abyss.
The canon hero from D1, the warrior, is referred to as the Dark Wanderer after his possession by Diablo's soul-stone.
Adria is the witch from D1.
D3 beta:
The Dark Wanderer's name is Aiden. He is one of Leoric's sons.
Leah is Adria's daughter, Deckard adopted her at a young age. She refers to Deckard as uncle.
It is implied in the beta that Leah is Aiden's daughter. Adria "consoles him as best she can" after he takes in the soul-stone. This would imply that Leah is conceived after Aiden's possession. This means Leah is both the grand-daughter of Leoric and the last of the royal line, and that she is potentially Diablo's direct offspring.
According to Blizzard Belial, the lord of lies is the boss of act 2.
The color of the meteor impact is blue/teal. Not red like you would expect.
The meteor landed in the cathedral and awoke Leroic. Only people who managed to escape were Leah and Deckard. According to videos she was outside watching it.
Deckard is trapped inside, and one of the first quests is to rescue him.
Leah is the ONLY one near the impact site that was totally unharmed.

I was unable to determine the act 1 boss? Anyone? From what I hear Leoric is the halfway mark. But no reliable source on that.

Additionally Tyreal is codenamed Bridle. What's with the horse/pony references?
Diablo is a boss. He is both referred to as the Lord of Terror, and The Prime Evil. Speculatively he is the boss of act 4.
There will be a 5th and 6th act, speculation is that they are reserved for expansions. No zones exist for them. Level cap titles go up to 100, further proof of expansions.
Act 4 includes both Heaven and Hell. Silver spire is the name of the heavenly areas.
My Speculation:
Heaven and Hell will both play a part in this, and I think the Meteor had to have come from heaven. Hence the soft blue color palette as apposed to demonic red. Or even fiery as one might expect. It seems as if it were aimed at Leah or Deckard. Possibly something else that we don't know. It seems that Leoric awakening was only a side-effect of the impact.
With the implications that Leah could literally be Diablo's daughter it would make sense that the meteor could have been an attempt to kill her.
A question for those who know a lot about the lore, if a prime evil has offspring, what does that mean for them? Theoretically could Diablo have gone to his offspring's body instead of the Black Abyss when his soul-stone was destroyed. Leah would have been just born or still in the womb at this time. Also if you look at the diablo 3 street art from Germany, Daiblo appears more feminine than before. If a prime evil is in the Black Abyss can a new entity claim their title? Can there be a new Diablo, Lord of Terror, or does it need to be the original soul/essence?

We know Deckard Cain spent the 20 years between Diablo 2 and Diablo 3 researching prophecies on the return of the Prime Evils. If I was raising a girl with some of Diablo's essence in her and I was the last member of the Horadrim order, I think I'd like to keep her close, you know, just in case she could be a gateway to a Prime Evil.

The star could be Imperius trying to kill Leah before Diablo can use her. It could be Tyrael coming to warn the mortals of the imminent danger. I'm leaning more towards Tyrael. The star crater in the Beta has blue/white glowing edges, and Imperius seems associated with red/orange fire in concept art.

Though there are countless story speculations/-summaries over the net, I really like this one. I myself have left the lore speculations for others, because of the headache it was giving me :).
But still I'd like to add that it seems that Adria is captured (and possibly possessed) in act 2 and then she betrayes us in act 3 (Adria--Azmodan).
I'm too tired to dig out the datamined lines I'm referring now, but I'll edit my post tomorrow.

A lot of this is exactly what I was thinking. I hadn't thought about the three merging though. If it were Tyrael coming why didn't he show himself, or maybe that's right after the Leoric quest and why the beta stops there. They stated that the beta ends when it does to avoid a major plot point.

The theories here are actually quite good:D However, after the Worldstone was destroyed, are the Prime Evils still required to enter sanctuary through host bodies? I thought the destruction of the Worldstone meant that demons could enter Sanctuary freely, in their true form. If this were the case, Diablo wouldnt really need Leah for anything.
Correct me if I am wrong! I do not remember what litterature and/or speculation I used to come to this conclusion.
Yes you're correct, Primes would no longer need a human host IF they were "alive and well" in Hell. But as you probably know they are in the Black Abyss, from which we know only a little.
My speculation is that when Leah was conceived, Diablo left a small piece of himself in leah. Be this unintended or planned by Diablo, I don't know.
It just seems weird. Diablo using Leah to somehow getting out of the Black Abyss I can understand. Using Leah as a host body just to wreak havoc on Sanctuary I would not understand. If you can enter Sanctuary in your true form, you should. If you are stuck in the Black Abyss, you should try to get out of there, and then enter Sanctuary in your true form. If Leah plays a part in this, I would understand possessing her.

I'm still not buying into this whole "possess Leah" theorizing, mainly because it just doesn't seem needed at this point.
That being said, Leah is obviously paramount to the story, even more so now that we know her lineage. But it could just as well mean that she will be instrumental in defeating Diablo, thereby finally redeeming her entire family who have been plagued by the Lord of Terror for generations.
PlugY for Diablo II allows you to reset skills and stats, transfer items between characters in singleplayer, obtain all ladder runewords and do all Uberquests while offline. It is the only way to do all of the above. Please use it.

That being said, Leah is obviously paramount to the story, even more so now that we know her lineage. But it could just as well mean that she will be instrumental in defeating Diablo, thereby finally redeeming her entire family who have been plagued by the Lord of Terror for generations.
At the very least we seem to be getting ample evidence that killing demons permanently isn't as easy as just bleeding them to death.

Because she betrays us apparently. And Diablo is female. That is the extent of my evidence. Less likely than Leablo I think, BUT TOTES A POSSIBILITY.
