The runed effect would be better for burst damage but u lose the power of the DoT. Does anyone know if the poison damage over time stacks or whether spamming them would restart the poison counter. The spider theory sounds effective however I dont think its my style of gaming.
it doesn't stack, but you can weave spells between it while the dot is ticking. It's actually probably the optional use of the spell.
killing speed increased wayyyy more in the beginning of the beta once psn dart was runed.

In order for delayed abilities to be more optimal than a straight damage option they need to provide more total damage over their duration than straight active use.
Unruned Poison Dart does 100% + 80% over 2 seconds. Throw in 2 seconds of Crimson Firebats at 170% Weapon Damage. 180 + 340 = 520% Weapon Damage.
Splinters Poison Dart x3 = 630% weapon damage.
Therefore Unruned Poison Dart is not a candidate for supportive DPS.
Abilities currently true supportive dps: non-Alabaster Haunt, Corpse Spiders, Grasp of the Dead, Alabaster Spirit Barrage, Crimson or Obsidian Locust Swarm, and of course Dogs or Gargantuan.

But poison dart, throw jumping spiders, cast another spell is more damage.
180% + 240% (Not probable, more likely 180%ish) + 425% eels = 845%
Obviously you can't use eels every time through, but you can weave other skills in as well.
It doesn't have to always be perfect. But it would allow you to use other runes, like the mana regen one. It also means you aren't spamming the same skill always.

Under that same scenario though is Splinter Dart 210 + 240 + 425 = 875%
In order to be a supportive dps you need to do more damage than can be done with a single use than any other ability that could be cast in its place, unruned poison dart doesnt apply because just using splinters straight outdamages it.

As a note, splinter dart has a longer cast time than unruned.
