Do you think its going to work the same way in Diablo 3? I'm excited to find more about the lore than I did before.
I imagined of having 2 ~ 3 bosses that are related to one another, so you kill one and move on to the next one etc...
But of course, the last one would be the hardest
This would be fun since there will be more bosses to kill.
Not just 4 main bosses from act 1 ~ 4 combined.

Well... There could be a large number of "mini" bosses in each act leading up to the final boss. There could also be multiple versions of the final boss, like once you kill one version the boss shifts into another. So you could have lots of different versions of bosses per act. So what your really asking is... should there be more than one very hard boss to end a certain chapter within an act, before the final hard boss to end the act?
Edit: I forgot to give my opinion... I think it would be fun to have lots of mini bosses during the act. Plus some optional aswell as mandatory side stories that end with large bosses to unlock certain things or a chance for special gear. So you could choose to rush through straight to the end, or take a bit more of a varied path with extra story.
I think there will be more than 1 difficult boss per act. It's expected to have at least one per act, as a challenge and as a way of sure fire loot of some sort. Since in all reality a completely different group of people have made this game than D1 and D2, I feel they'll put their own twists on boss to act composition instead of the 1/1 there was in D2. Between the mini bosses which look amazing so far, and elite bosses I have high hopes for a lot of epic challenges.

I'm like 99% sure there are more bosses in each act, but I do not know how mandatory they are. We've seen a few boss fights already and I'm pretty sure none of them are act bosses, because Blizzard probably wouldn't have shown us those. I remember a blue post also saying that the Skeleton King was not an act boss, so I know for sure that one isn't.
I'd like at least two important, mandatory bosses per act. But of course, the "act boss" will obviously be a lot stronger. But it'd be nice if the quest related bosses were tougher like act bosses.
The super unique monsters in D2 sort of had a bossy feel to them, like blood raven and griswold etc. I have the feeling that D3 will have these sort of mini-bosses but they will be much much more epic such as siegebreaker and the skeleton king. These battle are already pretty awesome and its hard to imagine what the final act bosses will be like
I like the idea of more than one boss, but I also like the idea of boss group (like the ancients) where you have to battle more than one aggressor. This open the door for a lot more interest and variety among the boss options, although I might be leery off a boss mob.

Well...I think it really depends on how they distribute them honestly. But something tells me we might be seeing more than 1 boss per act.
My main reason for that is based upon the first gameplay trailer. The siege breaker is clearly nowhere near the sophistication and power of a "real" boss like Diablo or the lessers such as Azmodan or Belial. Yet, he's freakin' brutal!
Did you see that?!?!? I don't care how insignificant he is, that is a BOSS move!lol. And thats just 1 monster. We could come in conflict with countless demons similar in ruthless vigor. And then there's guys like King Leoric, Bartuc, e.t.c. The list goes on really. None of those characters are really big names in comparison to Diablo or the higher ups but they would certainly be worthy bosses.
So, I say it all depends on who Blizz wants us to fight. They set up a really nice pyramid of power for us to climb. Where, even on the bottom the bosses are no laughing matter.

1 Act Boss only in Diablo 3 but the super-unique monsters will feel like act bosses. King Leoric is "just" a super unique with his own dungeon. Same should be with the Siegebreaker. Both of those are act 1. Imagine what they didn't show us yet. I feel certain it's going to be very impressive.
They have said their will be a final conflict concluding the act and transitioning to a new one.
As for bosses, I guess it all depends on what you mean by "boss" because I felt, as mentioned by others, that the countess, the raven, griswold, summoner, ... were all bosses. Sure, they were just uniques but there was some story to them and they usually concluded a quest.
As for D3, again as stated numerous times in this thread, yes there will be many bosses per act. The skeleton king and thousand pounder are proof of this. However, there will still be one epic boss fight at the end of each act.
After seeing leoric and thousand pounder, I am very excited to see just how awesome act boss fights are going to be. If thousand pounder can pick you up and bite your head off, just imagine what Diablo will be able to do to you.

The other bosses would be mini bosses, and would be less challenging than the final boss, but still a challenge nonetheless.
