Having a lot of information about the game now I have been thinking about what I will most likely occupy myself with during the endgame. As with d2 the endgame will be very rich with things such as the following:
PVM
PVP
Trading
Crafting
Trying different builds
MF'ing
In d2 I spent most of my time trading and playing an economy type game during the endgame. The trading in d3 should be greatly improved over the very primitive spam channel trading in d2 ( pre-d2jsp days) so I have a feeling that I will once again be a trader more than a player. Although of course it is difficult to predict this without having played the game, for all I know I may LOVE the new PVP system and get hooked.
PvP will be my motivation, but seeking new items and searching for loot on my PvE characters will be the means to my end game. They mentioned that the gaining loot should be done by doing the most fun thing - makes sense, but to them that means co-op play mostly and less emphasis on eye-bleeding MF boss runs. That might not be a bad thing after all...
You won't see me wasting my time killing the same stuff over and over for loot. I can't even believe people want to go that way. Diablo 2 is a great game, but certainly not because of that. Hey guys, lets just farm for items all day. Really?
I finish Hell, reach the level cap if its not stupidly time consuming for no reason like in D2, I do any end game stuff or events they have in store, and then I stop; next character. Don't care for PvP.
Its all about will they have anything decent to do. This isn't supposed to be a MMO, after all.
Heh, different strokes for different folks. As they mentioned in the Q&A panel, they're going to try to cater to broader interests. I certainly don't enjoy repetitive farming, but I do enjoy creative and fun ways of building my gear and wealth. Let's hope they can deliver on that (crafting materials, co-op bonuses, rare generated content, etc).
I got a feeling that we will all have a fruitful endgame consisting of all the above mentioned and more (events and such), despite what some say they won't do.
Basically, PVM with friends and constantly starting new chars wih different builds. Crafting sounds interesting so ill probably do that too. I will PVP sometimes for fun. Trading and getting richer will be my main goal at the first couple of months so i can afford new characters and gear.
It all kind of depends on what end game turns out to be upon release. What I'm really hoping for is a continual string of events that level-capped players can participate in. In keeping with the Diablo universe, I imagine these events to be somewhat random.
I started a new thread to get people's ideas for in-game events, seems like a worthy topic. Link at the bottom.
I find rote repetition and endless grinding profoundly depressing, and really hope Blizzard works the mechanics well enough so we won't have to flat boss-grind. If we do, I'll probably lean towards trading and PvP. I loves me some PvP so that'll really be my gear-up motivation. FTR, I was sad to find that we can't gank fools in-game, but then I'm a heartless sumbitch.

Different builds, crafting, and mfing. Maybe PvP, but that'll be something I look into after I beat the campaign.
I hate the way you cling to ignorance and pass it off as innocence
Yeah, I'll do that...
definitely!!
hmm, this too
aaaand this
not this....psych
can you take a guess?
I'm doing it all!!
PVM
Crafting - I always loved crafting.
Playing each character at least 10 times if not more (Hopefully different builds)
Hopefully when I get bored of D3 there will be mods for D3
I will likely assist in the making of D3 mods.

I saw somewhere where a pally used really poor items to beat uber trist.
After beating the game and as of right now, I dont know what i will do end game.
But i know that it will be challenging to try and beat the game with all blue lvl 20 items
