Does anyone know if the game is going to have a day and night cycle throughout all the levels? I was thinking that a certain level will be all night and others will be daylight.
No. Personally I hate day and night in games. Keep the towns daytime to give them a "warm" and "safe" feeling and make the scariest, grungiest areas of the map dark and at night.
Well, when you enter the towns at night, it should be lit by other sources of lights like torches or lanterns around the homes.
I know that some of the screenshots you see nights, which are amazing, but am wondering if they cycle.
I hope the nights are dark, as in, you can only see realistically based on the moon and sky above you and what conditions they are. For example, Full moon vs no moon, cloudy vs clear, stuff like that.
As for the weather effects, I hope they feel... epic. Like, real bad thunderstorm with wind and thunderous lightning, vs the weather in wow, which is pathetic. I realize in wow they have to consider the type of game it is (mmo vs 8 to 16 players per game) but wow's weather was/is awful in my opinion.
the day/night cycle should have an impact on the gameplay instead of just visual, such as certain monsters only spawn at night or certain time of the day, monsters gain hp faster at night and drop better items, and there should be items that allow players to change the weather and time

Maybe they could make areas that flood when it rains there?

I dont think that would work, because if it floods in certain areas there could be a boss, chest or something else important that we could miss, unless of course its a water boss or you could swim
It could work by making places that flood.
I don't think Blizzard would stick a Boss in a flood area.
They could do it like this.
A place floods as you're fighting through it... say a dungeon and you're underground and the roof caves in and it progresses as you make your way through the dungeon.
When you stop.. the flood would have to stop to prevent the character from dying tho.
There are many ways this can work.

Day/Night cycles are a must because they just add realism to the game. My suggestion would to literally have a cycle of days and nights(i.e. have the day correspond with the real world a full 24 hour day). Having a full 24 hour day would obviously be much more engaging being that you would get an experience of just exactly how long you've been playing and instead of a simple two light cycle, switch it up, have a twilight, dusk, dawn morning, night and adjust with colors as well. Instead of just having a simple yellowish white light shining on you make it a bright yellow and on some days have the sunset last for around 1 hour and change the colors up to a pinkish red or have the moon completely blocked by clouds and run around in near complete darkness. These are just some of the many things that Blizzard can do to spruce up the game and make it lively. I know i want to feel like im living in the world of Sanctuary

It only makes sense.
There were a lot of different cycles other than night in Diablo II tho.
I don't think they spanned the entire game, but they were there in some areas.

Day/Night isn't nescessary, depend where u are. Cave and monsatery are always dark anyway.
